LECTIN FROM CONCANAVALIN A TYPE VI - Introduction

A plant lectin (1ectin) extracted from grains such as concanavalin and pea. It is a tetrameric globulin with a molecular weight of 102000. Each subunit contains 237 amino acid residues, has a molecular weight of 25500, binds one Ca2 + and one Mn2 +, and contains a sugar binding site. It can specifically bind to α-mannose and α-glucose (on cell membrane glycoprotein), and the binding site is required to be the C- 3/C-4 and C- 6 in the six-membed ring of α-D-mannose or α-D-glucopyranose unsubstituted hydroxyl. It can have the greatest affinity with mannodisaccharides and mannotriosaccharides connected by α-1,2 glycosidic bonds. It is a cell-promoting mitogen (mito-gen), which mainly stimulates T lymphocytes, aggregates red blood cells and animal sperm, inhibits tumor cell movement, and prolongs the survival time of heterologous transplantation. Concanavalin A has no blood type specificity, but has affinity for terminal α-D-mannose and α-D-glucosyl residues. The presence of Ca2 + and Mn2 + ions is required to show activity. Concanavalin A dissociates to dimer at pH 6.5 or lower. When the pH is between 5.8 and 7.0, concanavalin A is present as a tetramer; when the pH is higher than 7.0, a higher polymer is formed. Depending on the degree of polymerization, concanavalin A may show mitogenic activity. Succinylation can produce an active dimer that can maintain the dimer form when the pH is higher than 5.6.
